I'd say The Coon isn't as dedicated to that objective beyond his first episode. All villains or anti-heroes who tend to throw common sense out the window when an opportunity to defeat their rival presents itself. I'd say a similar situation might be Doom and Reed Richards, or (going outside of superhero comics) Yugi vs Kaiba, Dio vs Jonathan, or (early series) Vegeta vs Goku. I think The Coon manages to get the better of Mysterion a couple times through underhanded means. While I think it's highly accurate to say they're each others nemesis (due to opposing ideologies, one being selfless and the other selfish, and openly hating one another) I wouldn't necessarily call Mysterion The Coon's Kryptonite. It's how he's able to mess with the entire Fractured But Whole plot to take over the town, make it Christmas everyday, and even get away relatively unscathed despite being openly known as a scummy anti-hero douchebag responsible for the town's problems. The Coon is more like Batman in abusing his mother's money to fund equipment (such as his razor sharp claws) to deal savage damage, however his real "power" comes in Cartman's unique and infamous ability to manipulate people and situations to get what he wants. Mysterion has an actual superpower of endless rebirth, martial arts skills and focuses on being unseen. But on paper, they're pretty neck and neck. Ideologically and popularity-wise Mysterion certainly bests The Coon.
Doesn't The Coon beat Mysterion during The Coon (episode) forcing him to unmask, and again during Civil War 2.0? Granted Mysterion defeats The Coon multiple times before The Coon's (episode) climax by surpassing him in crimefighting, and would have defeated him in Coon vs Coon & Friends if Mintberry Crunch hadn't usurped the victory.
